What the record says
Semaglutide: two large trials, and what they were testing
STEP 1 randomised 1 961 adults without diabetes to once-weekly semaglutide or placebo for 68 weeks. Mean body weight changed −14.9% against −2.4% on placebo, an estimated difference of −12.4 percentage points.
N Engl J Med 2021;384:989–1002 ↗SELECT enrolled 17 604 adults with overweight or obesity and established cardiovascular disease but no diabetes. A primary cardiovascular event occurred in 6.5% on semaglutide against 8.0% on placebo (hazard ratio 0.80; 95% CI 0.72–0.90) over a mean follow-up of 39.8 months.
N Engl J Med 2023;389:2221–2232 ↗Both are prescription trials on a fixed weekly schedule in a clinical setting. They measured the approved drug, not a compounded research vial.
